摘要
观察糖尿病患者血栓前状态若干相关分子标志物即血浆内皮素- 1(ET- 1) 、血小板α- 颗粒膜蛋白(GMP-140)、组织纤溶酶原激活物(tPA) 及其抑制物(PAI- 1) 的变化。方法:分别采用放免法和发色底物显色法,检测了20 例正常人和40 例Ⅱ型糖尿病患者(22 例无微血管病变,18 例有微血管病变)的ET-1 、GMP-140 、tPA及PAI- 1 的变化。结果:糖尿病患者的ET- 1、GMP- 140 、PAI- 1 显著高于正常对照组,tPA 则明显下降,伴微血管病变者比无微血管病变者变化更明显,且ET- 1 与GMP- 140 、PAI- 1 呈显著正相关,而与tPA 呈显著负相关。结论:上述血栓前状态若干相关分子标志物的测定对糖尿病微血管病变的早期诊断和病情分析有重要的临床价值。
To observe the changes of some molecular marks related to prethrombotic state such as the plasma endothelin-1(ET-1),platelet α -granule membrane protein(GMP -140 ),tissue-type plasminogen activator(tPA) and plasminogen activator inhibitor(PAI-1) in diabetics.Methods:ET-1,GMP -140 ,tPA and PAI-1 were measured by radioimmunoassay,and were developed respectively in 40 cases with typeⅡ diabetes mellitus(18 cases with microangiopathy,22 without microangiopathy) and in 20 control cases.Results:ET-1,GMP -140 and PAI-1 were significantly higher in the diabetic patients than those in the controls,whereas tPA was obviously lower.The changes of the above indexes were more apparent in the patients with microangiopathy than those in the patients without microangiopathy.There was a significant positive correlation between plasma ET-1 and GMP -140 ,PAI-1,whereas there was an obviously negative correlation between ET-1 and tPA.Conclusions:The measurement of the above molecular marks related to prethrombotic state has an important clinical value in the early diagnosis and analysis of diabetic microangiopathy.
