摘要
目的:建立大鼠急性放射性肝损伤模型.方法:雌性SD大鼠10只为正常阴性对照组,12只为照射组。对照组为正常饲养4周;照射组大鼠肝脏右叶均接受6兆伏(MV)-X线源皮照射技术,源皮距离(SSD)为80cm,照射深度为2cm,照射速度(吸收剂量率)为300cGy/min,单次照射总量为20Gy。每周观察大鼠的一般生活状态;4周后大鼠眼内眦静脉取血,检测血清谷丙转氨酶(ALT)、血清谷草转氨酶(AST)水平;普通光学显微镜下观察肝脏组织的病理形态学变化。结果:大鼠肝右叶接受单次辐射20Gy后2周,其一般生活状态差,II放射性皮炎,4用后大鼠存活率为91.7%(11/12);显微镜下大鼠肝脏右叶发生不同程度的损伤,主要表现为汇管区炎性细胞浸润、肝血窭水肿;照射组大鼠血清ALT为(44.00±11.95)U/L,高于对照组(26.00±9.63)U/L,t=2.8725,P=0.0166;照射组大鼠血AST为(108.50±20.69)U/L,高于对照组(69.17±8.84)U/U,t=4.2828,P=0.0016,P值均〈0.05,差异均具有统计学意义。结论:成功建立大鼠急性放射性肝损伤模型。
Objectlve: To establish acute radiation induced liver injury of rats. Methods: 10 female SD rats as normal negative control group and 12 female SD rats as irradiation group who were accepted 20Gy 6mv-X ray on the right lobe of the liver. Source skin distance(SSD) is 80em, the depth of irra- diation is 2cm, absorbed dose rate is 300cGy/min. The rats were bred in the same condition for 4 weeks and observed the following index.-a, the general conditions, b. Checking the ALT, AST level in serum, c. By the H--E staining, observing the pathomorphologieal change of liver. Results: Those rats that accepted 20Gy irradiation were in a poor general condition with II radiation dermatitis two weeks after irradition. Survival rate was 91.7%(11/12) in four weeks. In irradiation group, there are some injury of right lobe liver, such as edema of hepatic sinusoid and inflammation of hepatic header. Irradia- tion group's ALT (44.00±11.95U/L) and AST(108.50±20.69U/L was higher than that of in control group's (26.00+9.63U/L) and (69.17±8.84U/ L), respectively. Conclusions: Modeling of irradiation induced liver injury of rats were successful.
