摘要
目的:观察红藤对胃腺癌细胞MCG803细胞生长周期分布及细胞凋亡的影响。方法;采用MTT比色法观察10,20,40,80和160mg/L红藤处理48h对胃腺癌细胞MCG803细胞生长的影响,采用Gimesa染色技术、荧光染色技术进行MCGS03细胞凋亡检测,并用流式细胞仪检测40mg/L红藤处理24h对MCG803细胞周期分布的影响。结果:红藤可抑制胃腺癌细胞MCG803的增殖,红藤与细胞培养12小时即有少数MCG803细胞发生凋亡,随时间延长凋亡细胞数量逐渐增多,改变细胞周期分布,使G-0/G1期细胞比例增高,S期比例降低,并对MCG803细胞有促凋亡作用。结论:红藤可改变细胞周期分布,影响细胞周期调控蛋白表达,并可诱导MCG803细胞凋亡,从而抑制细胞增殖。
Objective: To study the effects of caulis sargentodoxae prescription(CSP) on cell proliferation and apoptosis on MCGS03 cells and to eluci- date its molecular mechanism. Methods: The proportion of the periodic tumor cells were altered by 10, 20, 40,80, 160 mg/L CSP treated 48 h using MTT and Sub--G1 curves were displayed at 40 mg/L CSP treated 24h by flow cytometry analysis. With Giemsa staining and fluorescence staining assay to detect the MCGS03 apoptosis. Results: CSP significantly Suppressed MCGS03 cell proliferation in a time-- and dose--dependent manner. The proportion of the periodic tumor ceils were altered by CSPo CSP decreased the proportion of cells in S phase and increased the proportion of cells in G0/G1 and/or G2/M phases of the cell cycle. Conclusion: These data support that CSP can induce apoptosis and alert cell cycle arrest of the Human Gliomaous Cells MCGS03
