摘要 目的:掌握外周血单个核细胞分离的原理和方法,形态学方法检测淋巴细胞增殖的原理及观察淋巴细胞转化后的形态学变化.方法:采用Ficoll密度梯度离心法分离外周血单个核细胞,用PHA培养所得的外周血单个有核细胞,采用形态计数法检测淋巴细胞的转化率。结果:本实验所得外周血单个棱细胞的分离纯度为92%,淋把细胞的转化率为85%。结论:Ficoll密度梯度离0法分离外周血单个棱细胞简单,所得细胞纯度高,PHA能有效地刺激淋巴细胞使其转化增殖,所用的形态学计数法简单容易掌握。 Objective To master the principles and method to separate PBMC in peripheral blood, as well as the morphological method of detecting the multiplication of the lymphocyte, at the mean time, observing the change of lymphocyte in morphology. Method To separate the PBMC in peripheral blood by means of Ficoll density gradient centrifugation method," experiment with the PBMC cultivating in PHA, and get the transformation rate of lymphocyte with the method of morphological counting. Result The separation purity of PBMC getting in this egperiment is 92% and the transformation rate of lymphocyte is 85%. Conclusion It is a simple way to get PBMC in peripheral blood separated with the F'icoll density gradient centrifugation method, and the separation purity is high. PHA can stimulate the transformation and cultivation of lymphocyte efficiently. What's more, the method of morphological counting which we use is very easy to master.
