简易HPCE法分离检测扑尔敏对映异构体

摘要 采用高效毛细管电泳(HPCE)法分离扑尔敏手性异构体,建立了简易HPCE法分离检测扑尔敏对映体含量的方法.在国产仪器上,以未涂层石英毛细管柱(75μm)、pH2.5、Tris-磷酸缓冲液(40 mmol/L)、分离电压25 kV、进样时间1 min、检测波长214 nm、温度20℃条件下扑尔敏两对映体实现了基线分离,分离度可达10.64.扑尔敏消旋体浓度在0.001~0.10 mg/mL范围内,峰高、峰面积与浓度呈线性关系,峰面积线性关系好于峰高,两峰峰面积线性相关系数r可达0.99883和0.99128,前峰的峰高及峰面积精密度实验表明,RSD(5次)分别为7.97%和8.29%.在未构建其他手性环境的条件下,该方法使扑尔敏两对映异构体达到快速分离,据此建立的测定方法可用于实际样品的含量测定. A simple method of enantiomorph separation and determination of chlorpheniramine by high performance capillary electrophoresis was investigated.The chlorpheniramine enantiomers were well separated in the uncoated fused silica capillary column(75 μm) under the following conditions:pH2.5,Tris-phosphate buffer 40 mmol/L,separation voltage of 25 kV,20 ℃,injection time is 1 min,detection wavelength λ of 214 nm.A good linear relationship was obtained between the peak height or peak area and chlorpheniramine racemoid concentration the range of 0.001~0.10 mg/mL,the linear relationship coefficients(r) of peak areas is 0.99883 and 0.99128 for the first and second repectively and the relative standard deviations RSD(5 times) of the first peak height and peak area is 7.97 % and 8.29 %.
