摘要
对大球盖菇(Stropharia rugoso-annulata)原生质体制备的条件进行了研究,并确定了最佳条件组合:选用15 g/L的纤维素酶+15 g/L的蜗牛酶的混合酶来酶解菌丝,0.6 mol/L的甘露醇作为渗透压稳定剂,菌龄为3 d,酶解温度30℃,酶解时间2.5 h,在此条件下制得的原生质体产量最高达1.70×107mL-1.本实验还对紫外线对大球盖菇原生质体的诱变效应进行了探究,选取35 s为最佳诱变照射时间,致死率是73.33%.
Conditions of preparating protoplast of Stropharia rugoso-annulata were studied. The result shows that the highest production rate of protoplast(1.70 × 10^7 mL^-1) can be obtained on these conditions. 3-day-old mecia, 15 g/L cellulase and 15 g/L snailase admixture, dissolution in 0. 6 moi/L mannitol, resolved at 30 ℃ for 2.5 h. Ultraviolet radiation inducing S. rugoso-annulata protplast was researched too. The conclusion is that 35 s is the irradiation time, deadly ratio is 73.33%.
