重组人Ⅵ型胶原蛋白多肽对长波紫外线辐射的人皮肤成纤维细胞损伤的保护作用

Protective Effect of Recombinant Human Collagen Peptide Type Ⅵ on Human Skin Fibroblast Lesion Caused by Long-wave Ultraviolet

摘要 目的探讨重组人Ⅵ型胶原蛋白多肽(CP6)对长波紫外线(Ultraviolet A,UVA)辐射的人皮肤成纤维细胞(Humanskin fibroblast,HSF)损伤的保护作用。方法 MTT法检测不同剂量UVA辐射对HSF细胞活力的影响,并采用MTT法、An-nexin V/PI流式细胞术和TUNEL法分别检测CP6对UVA辐射的HSF损伤的保护作用。结果辐射强度为5和10 J/cm2的UVA可造成HSF活力降低;与UVA对照组相比,UVA+4 mg/ml CP6、UVA+6 mg/ml CP6和UVA+8 mg/ml CP6组A570值均升高(P<0.05),细胞凋亡率显著下降(P<0.05)。结论紫外线可造成体外培养的HSF损伤,一定浓度的CP6可提高辐射细胞的存活率,减少细胞凋亡,具有保护作用,其机制可能与其抗氧化、增强细胞活力有关。 Objective To investigate the protective effect of recombinant human collagen peptide typeⅥ(CP6) on human skin fibroblast(HSF) lesion caused by long-wave ultraviolet(Ultraviolet A,UVA).Methods The effect of UVA at various dosages on activity of HSFs was analyzed by MTT method,and the protective effect of CP6 on UVA-caused HSF lesion by MTT,Annexin Ⅴ/PI flow cytometry and TUNEL method.Results UVA at dosages of 5 and 10 J/cm2 decreased the activity of HSF.The A570 values in UVA + 4 mg/ml CP6,UVA + 6 mg/ml CP6 and UVA + 8 mg/ml CP6 groups were significantly higher,while the cell apoptosis rates were significantly lower,than that in UVA control group(both P 〈 0.05).Conclusion UVA caused the lesion of HSF cultured in vitro.The CP6 at a certain concentration increased the survival rate and decreased the apoptosis of irradiated cells,of which the protective effect might be associated with antioxidation and increasing cell activity.
