摘要
以含有r - 谷氨酰半胱氨酸合成酶( GSHI) 和谷胱甘肽合成酶( GSHⅡ) 的S-cerevisiaeemulsefier- E472c 酵母菌株为酶源, 以谷氨酸、半胱氨酸、甘氨酸为底物, 酶法制备谷胱甘肽(GSH) 工艺的研究。采用了直接高效液相色谱法检测酶促反应中GSH 的含量, 建立了酶促反应的最佳反应条件。在优化反应条件下, 对底物半胱氨酸的转化率为80 % ,经过分离纯化回收率为83 % ,总产率为66-4 % 。结果表明, 该法制备GSH 成本低, 工艺过程简单, 能够适用于GSH 的工业化生产。
A simple and economical method of enzymatic synthesis of Glutathione was developed Glutathione was produced by means of the biotransformation of L Glutamic acid L Cysteine and Glycine in the presence of yeast cell of S cerevisiae emulsefier E 472c The analytical method of determine GSH and GSSG with HPLC was founded and a series reaction conditions were optimized Under this condition, the transformation rate can approach 80% with a mole yield of 82%.
