摘要
目的:探讨鲨鱼软骨粉(SCP)联合“425”制剂诱导的抗血吸虫病及抗血吸虫性纤维化效应。方法:BALB/c小鼠感染日本血吸虫后4w,灌服SCP合剂,服药后3w剖杀;胸主动脉灌注冲虫,计算减虫率、减卵率;制备肝石蜡连续切片,测定肉芽肿大小及观察纤维化程度;ELISA检测血清IgG和IgG_1抗体。结果:SCP合剂大剂量(90mg)组1小鼠减虫率50%,显著高于SCP合剂小剂量(70mg)组Ⅱ的33.3%、单用SCP组Ⅲ的23.3%和单用“425”制剂组Ⅳ的32.0%(P<0.05);组Ⅰ雌虫子宫虫卵数(平均75.3±18.51)显著低于组Ⅱ(102.2±11.91)、组Ⅲ(108.5±10.50)、组Ⅳ(115.8±10.83)和感染对照组(139.0±27.7)者(P<0.05);组Ⅰ小鼠肝虫卵减少率为81.93%,显著低于组Ⅱ~Ⅳ的47.19%、44.4%和28.15%(P<0.01);组Ⅰ~Ⅳ小鼠肝虫卯肉芽肿(SEG)大小均显著小于感染对照组者(P<0.001),组I小鼠肝SEG大小显著小于组Ⅱ、Ⅲ和Ⅳ者(P<0.05),而组Ⅱ、Ⅲ、Ⅳ小鼠SEG大小间无显著性差异(P>0.05);组Ⅰ、Ⅲ小鼠肝SEG内纤维化消失,而组Ⅱ、Ⅳ小鼠肝SEG内可见纤维组织,但感染对照组SEG内显示重度纤维化。用药后3w组Ⅰ、Ⅱ和Ⅳ小鼠血清IgG和IgG_1水平均显著高于组Ⅲ和组Ⅴ者(P<0.01),而前3组间和后2组间抗体水平无显著性差异。结论:SCP合剂对日本血吸虫具有明显的减虫。
Aim:Further to probe the effects of anti-S. japonicum egg granuloma and hepatic fibrosis in infected mice induced by shark cartilage powder (SCP) cooperating '425' preparation. Methods: The BALB/c mice infected with cer-cariae of S. japonicum were orally administered using SCP mixture (90mg in group Ⅰ , 70mg in group Ⅱ ), SCP only (group Ⅲ), '425' preparation only (group Ⅳ ) and normal saline as control group Ⅴ at 4w post-infection and were killed at 3rd wpost-treatment. The number of adults and eggs in liver was counted respectively, and the extent of hepatic schistosome fibrosis and the mean sizes of schistosome egg granuloma (SEG) were observed by morphometric examination of liver tissue. The specific antibody IgG and its subclass IgG1 levels were detected by ELISA. Results:The reduction rate of adults worms in group Ⅰ (50%) was significantly higher than that in group Ⅱ(33. 3%). group Ⅲ(23. 3%) and group Ⅳ (32.0%) (P<0. 05). The number of eggs in the uterus of female adults in group Ⅰ(75. 3± 18. 51) was significantly lower than that in group Ⅱ(102.2±11. 91), Ⅲ(108. 5±10. 50).Ⅳ(115. 8± 18. 51) and Ⅴ(139.0±27. 7) respectively (P<0. 05). The reduction rate of hepatic eggs in group Ⅰ (81. 93%) was markedly higher than that in other groups ( Ⅱ-Ⅳ:47. 19% , 44. 4% and 28. 15% respectively) (P<0. 01). The mean sizes of SEG in group I . J . I and IV were significantly decreased than those in group Ⅴ ( (P<0. 05). and the mean sizes of SEG had not difference markedly among the groupⅡ. Ⅲ and Ⅳ , There was no fibrosis in the murine hepatic SEG of group Ⅰ andⅢ . and light fibrosis in group Ⅱ and Ⅳ , but serious fibrosis in group Ⅴ . The levels of IgG and IgG1 in sera of group Ⅰ. Ⅱ and Ⅳ were obviously higher than those in group Ⅲ andⅤ(P<0. 01). Conclusion: Shark cartilage powder mixture may play a certain role in killing S. japonicum and anti-embryo, and could strongly induce anti-pathological and anti-hepatic fibrosis effects and the effects of the larger dosage SCP mixture are better than those of the smaller one.
