EGFR、VEGF在非小细胞肺癌中的表达意义及与微血管密度之间的关系 被引量:16

Expression and significance of EGFR,VEGF in Non-Small-Cell lung cancer and the relationship of them with MVD

摘要 目的检测表皮生长因子受体、血管内皮生长因子在非小细胞肺癌(NSCLC)中的表达情况及NSCLC的微血管密度,探讨三者与NSCLC临床病理特征之间的关系及三者彼此的关系。方法采用免疫组化(S-P法)检测EGFR、VEGF在136例NSCLC及28例癌旁正常对照组织中的表达情况,通过CD34染色计数136例NSCLC及28例癌旁正常对照组织的MVD。结果 EGFR、VEGF在NSCLC组织中的阳性表达率及MVD明显高于癌旁正常对照组织。EGFR、VEGF的阳性表达及MVD均与NSCLC的TNM分期和有/无转移有关(P<0.01);VEGF的阳性表达与NSCLC的分化程度有关(P<0.05);三者均与NSCLC的组织学类型无关(P>0.05)。Spearman相关分析显示EGFR与VEGF在NSCLC中的阳性表达呈正相关(P<0.01);VEGF在NSCLC中的阳性表达与MVD呈正相关(P<0.01);EGFR在NSCLC中的阳性表达与MVD无明显相关性(P>0.05)。结论 EGFR、VEGF在NSCLC组织中高表达可促进肺癌的侵袭和转移。VEGF主要通过促进新生血管的形成,加速肺癌的侵袭和转移。 Objective To detect the expression of epidermal growth factor receptor(EGFR),vascular endothelial growth factor(VEGF) and microvessel density(MVD) in non-small cell lung cancer(NSCLC) tissue,then analyze the relationship of these indices with the clinicopathologic features of NSCLC and the correlation with each other.Methods The immunohistochemical S-P method was used to detect the expressions of EGFR,VEGF and MVD in 136 cases of NSCLC and 28 cases of para-cancerous normal lung tissues.Results The positive rates of EGFR and VEGF,as well as MVD in NSCLC were dramatically higher than that in the para-cancerous normal lung tissues(P0.01).The expression of EGFR,VEGF and MVD were significantly correlated with TNM stage and lymph node metastasis(P0.05),but were not correlated with the histological types of NSCLC(P0.05).The expression of VEGF was also correlated with grading of NSCLC(P0.05).Spearman correlation analysis indicated the expression of EGFR and VEGF was highly positive correlation in NSCLC(P0.01),VEGF expression was closely correlated with MVD(P0.01) but EGFR expression was not(P0.05).Conclusion The high expression of EGFR and VEGF in NSCLC plays important role in invasion and metastasis of NSCLC.VEGF accelerate the invasion and metastasis of NSCLC by augment the number of tumor's vessel.
