摘要
应用金黄地鼠胚胎细胞转化试验检测了氟化物的诱变致癌性。结果显示,1、4m m ol/L氟化钠(NaF)受试组与阴性对照存活约80d,未见转化发生。而2m m ol/LNaF处理组(SHEF2)细胞逐渐出现类似阳性对照MNNG 诱导的转化细胞特征;培养至110d 有转化集落形成;接种培养150d 的SHEF2 于裸鼠腹部皮下有纤维肉瘤形成。
The Syrian hamster embryo cultured cells were exposed to sodium fluoride(NaF)for 24 hours with concentrations of 1mmol/L,2mmol/L and 4mmol/L for cell transformation.A dose depended cytotoxity was observed.Moreover,the groups of 1mmol/L,4mmol/L and controlled were not transformed,which had a limited lifetime of about 80 days.Mass culture treated with 2mmol/L NaF(SHEF2)escaped senescence and gradually showed characters of transformed cell that is similar to those of positive controlled. SHEF2 cells proliferated rapidly with active function,and formed transformed clonies when cultured to 110 days.Incubation with agglutinin resulted in strong agglutination,furthermore,fibrosarcoma developed in nude mice inoculated with SHEF2 cells.All the phenomena suggested that malignant transformation of SHEF2 cells had occurred.
