14-3-3σ蛋白表达下降在鼻咽癌发病中的意义

摘要 目的探讨14-3-3σ在鼻咽癌(NPC)中的表达及其临床病理学意义。方法应用W estern b lot检测14-3-3σ在不同分化程度和不同转移潜能鼻咽癌细胞系中的表达;采用免疫组织化学S-P法检测98例原发NPC组织、30例正常鼻咽黏膜上皮组织及20例颈淋巴结转移NPC组织中14-3-3σ蛋白的表达情况;采用SPSS 13.0软件包对数据进行统计分析。结果 NPC细胞中,14-3-3σ表达与分化及转移有关(P<0.01);14-3-3σ在原发NPC组织中的表达较正常鼻咽黏膜上皮组织明显下调(P<0.01);14-3-3σ在颈淋巴结转移NPC中的表达水平较原发NPC明显下调(P<0.01);98例鼻咽癌组织标本中,14-3-3σ的表达水平与NPC的病理分化程度、临床分期、复发及局部淋巴结和远处转移有关,14-3-3σ低表达的肿瘤较高表达的肿瘤有更晚的临床分期、更容易复发、更容易发生局部淋巴结和远处转移(P<0.05)。结论 14-3-3σ在鼻咽癌中表达降低可能与其发生、发展有关,有望成为NPC分化的标志物及潜在的治疗靶标。 Objective To investigate the expression of 14-3-3σ in nasopharyngeal carcinoma(NPC) as well as its correlation with clinical pathology.Methods The protein expression of 14-3-3σ in some NPC cell lines was detected using Western blotting.Immunohistochemistry was performed to detect the expression of 14-3-3σ in 98 cases of human primary NPC tissue specimens,30 cases of normal nasopharyngal epithelial tissue(NNET),and 20 cases of cervical lymph node metastases NPC(LMNPC).The correlations of 14-3-3σ expression level with clinicopathologic features and clinical outcomes were evaluated with SPSS13.0 software package for non-parametric analysis.Results 14-3-3σ expression was correlated to differentiation and metastasis of NPC(P0.01).14-3-3σ was significantly down-regulated in NPC versus NNET(P0.05).The expression level of 14-3-3σ in primary NPC was significantly higher than that in LMNPC(P0.01).14-3-3σ down-regulation was correlated with poor histologic type/grade,advanced clinical stage,recurrence,regional lymphonode and distant metastasis(P0.01).Tumors with 14-3-3σ down-regulation tended to have a more advanced clinical stage,more frequent recurrences,more lymph node and distant metastasis(P0.05 or 0.01).Conclusion The results suggest that decreased expression of 14-3-3σ contributes to cancerous progression,and 14-3-3σ may be potentially used as a differentiation biomarker and a therapeutic target for NPC.
