摘要 目的:了解兔源性透明质酸刺激因子(hyaluronicacidstimulatingfactor,HASF)对人增生性瘢痕成纤维细胞生长增殖的影响.方法:将HASF作用于体外培养的增生性瘢痕成纤维细胞,测定3H-TdR参入量、MTT反应A值及上清液中透明质酸(HA)的含量.结果:1×10-3~1×10-2g/LHASF可剂量依赖性地使细胞3H-TdR参入量及MTT反应A值降低,HA含量明显增加(P<0.01).结论:兔源性HASF在体外能促进人增生性瘢痕成纤维细胞合成HA,对其生长增殖有明显的抑制作用;这种抑制作用可能是通过HASF刺激细胞合成HA来实现的. AIM: To study the influences of hyaluronic acid stimulating factor (HASF) on the growth and proliferation of scar derived fibroblasts. METHODS: 3H TdR incorporation and MTT OD value was detected and the content of HA was determined with radioimmunology after the HASF was added to the monolayer culture system of scar derived fibroblasts. RESULTS: HASF at 1×10 -3 ~1×10 -2 g/L could decrease the 3H TdR incorporation and the MTT OD value in a dose dependent manner, and apparently increase the content of HA( P <0.01). CONCLUSION: HASF can promote scar derived fibroblasts to synthesize HA and inhibit its growth and proliferation in the monolayer culture system. This inhabitation may be caused by the increase of HA, which was synthesized by fibroblasts and stimulated by HASF.
