摘要
依据生物的不对称降解或同化作用机理,在特定的培养条件下培养假丝酵母(Candida.maltosa),产生对L-丙氨酸具有强氧化能力的氨基酸氧化酶,不对称催化氧化底物DL-丙氨酸中的左旋异构体脱去氨基分解,从而制备D-丙氨酸。研究了碳源、氮源、pH、温度对酶活力的影响,结论是选择反丁烯二酸为碳源,浓度为1.0%,有机氮源最佳浓度为:蛋白胨0.1%、牛肉膏0.4%、玉米浆0.6%,pH为6.0,转化温度为50℃时酶活力最佳。
Based on the asymmetric biological degradation or assimilation mechanism,the cultivation of the specific conditions training Candida(maltosa),produce Candida.To L-alanine has strong oxidation ability of amino acids,asymmetric catalytic oxidation oxidase substrate DL-alanine off the levorotatory isomers amino decomposed to preparation D-alanine.Research the carbon sources,nitrogen source,pH,the influence of temperature on the enzyme,conclusion is choice trans-butenedioic acid as the carbon sources,concentration of 1.0%,organic nitrogen source optimal concentration for: specially designed 0.1%,beef anointed 0.4%,corn plasma 0.6%,pH for 6.0,transfer temperature for 50 ℃when the enzyme activity is the best.
