摘要
目的观察健康人和不同类型乙肝患者PBMCs中Foxp3、Tim-3的表达和IFN-γ含量的变化情况,探讨CD4+CD25+调节性T细胞在乙肝慢性化中可能发挥的作用。方法采用RT-PCR检测不同类型乙肝患者和健康人外周血中Foxp3和Tim-3的表达情况。应用ELISA的方法检测血清中IFN-γ的含量。结果急性乙肝患者Foxp3的表达与健康组差异无统计学意义,但Tim-3的表达较健康组低(P<0.01),并且血清中IFN-γ的含量有所增加(P<0.05)。慢性乙肝患者Foxp3和Tim-3的表达均明显高于健康对照组(P<0.01),血清中IFN-γ的含量降低。结论慢性乙肝患者CD4+CD25+调节性T细胞活性有所增强,且Th1细胞活性和体液免疫均受到抑制,CD4+CD25+调节性T细胞在乙肝慢性化中起着一定的作用。
Objective To observe the expression of Foxp3 and Tim-3 and serum level of IFN-γ in PBMCs of hepatitis B patients and healthy individuals,and to explore the possible role of CD4+CD25+ regulatory T cells in the chronic procedure of hepatitis B.Methods The expression of Foxp3 and Tim-3 in PBMCs of hepatitis B patients and healthy individuals were determined by RT-PCR,and the serum level of IFN-γ was detected by ELISA.Results No statistically significant difference was found in the expression of Foxp3 between acute hepatitis B patients and healthy individuals(P0.01).As compared with healthy individuals,the expression of Foxp3 in acute hepatitis B patients was decreased(P0.01),moreover,the serum level of IFN-γ was increased(P0.05).The expression of Foxp3 and Tim-3 in chronic hepatitis B patients was significantly higher than that in healthy individuals(P0.01),but the serum level of IFN-γ was decreased.Conclusions In this study,we found that CD4+CD25+ regulatory T cells showed the stronger activity in chronic hepatitis B patients,which indicated they might play an important role in the chronic procedure of hepatitis B with suppressing Th1 and humoral response to hepatitis B virus.
