摘要
本研究应用人鼠游离胃平滑肌细胞和采用[35S]GTPγS结合法测定肌细胞的G蛋白,观察促胰液素舒张胃个沿肌细胞中的G蛋白的变化及其信号转导通路。结果显示:(1)促膝液素对胃体、胃窦平滑肌细胞均有舒张作用,促膝液素抗血清可阻断促腔激素对胃窦平滑肌细胞的舒张反应,Forskolin与cAMP抑制剂分别加强和抑制促胰液素的这一作用。(2)Gαs抗体可抑制促胰液素的舒张作用,促胰液素明显引起Gαs抗体与[35S]GTPγS结合的增加,表明胃平滑肌细胞中促胰液素受体与Gs蛋白相偶联,激活cAMP系统介导促胰液素舒张胃平滑肌细胞。
The aim of study was to elucidate the G protein involved in secretin-induced relaxation of gastric smooth muscle cell in rat. Gastric muscle cells were isolated by enzymatic digestion with collagenase.[35S]GTPγS-binding properties of G proteins were assesed in crude membranes of gastric muscle with or without secretin stimulation. Results:(1)Secretin relaxed smooth muscle cells of both gastric body and antrum. Secretin antiserum blocked the relaxation of secretin on antrum smooth muscle cells.Forskolin and cAMP inhibitor potentiated and weakened the effect of secretin, respectively. (2)The relaXation induced by secretin was blocked by Gαs-specific antibodies. Secretin caused a significantly increase in [35S]GTPγS binding to Gαs, but not to Gαq/11or Gαi-3. In conclusion, secretin activates a cAMP dependent pathway through Gs proteins in antrum
