HPLC法测定关苍术超临界萃取物中atractylenoideⅠ和atractylenoideⅢ的含量

摘要 [目的]建立关苍术超临界萃取物中atractylenoideⅠ和atractylenoideⅢ的含量测定方法.[方法]色谱柱为C18柱,以甲醇-乙腈-水(59.5∶0.5∶40.0)为流动相,流动速度为1.0mL/min,检测波长为220nm,柱温为28℃.[结果]atractylenoideⅠ和atractylenoideⅢ分别在进样量为70~560ng(r=0.999 7)和39~390ng(r=0.999 9)范围内呈良好的线性关系,平均回收率分别为100.80%和101.52%;RSD值分别为4.95和5.75.[结论]HPLC法灵敏、准确、重现性好,可作为关苍术超临界萃取物中atractylenoideⅠ和atractylenoideⅢ的含量测定方法. OBJECTIVE To establish a method of determining the content of of atractylenoide I and atractylenoide Ⅲ in supercritical fluid extracts (SFE) of the Rhizoma Atractylodis japonica. METHODS The high performance liquid chromatography chromatography (HPLC) was used with C18 column as chromatographic column, methanol acetonitrile water solution (59.5 : 0.5 : 40.0) as mobile phase, 1.0 mL/min as flow rate, 220 nm as the detective wavelength, and 28 ℃ as column temperature. RESULTS The atractylenoide I and atractylenoide Ⅲ had favorable linear relation as the sampling volume of 70 560 ng (r = 0. 999 7) and 39-390 ng(r = 0. 999 9), the average recovery was 100.80% and 101.52%, and RSD was 4.95 and 5.75, respectively. CONCLUSION The HPLC is sensitive, accurate, and repeatability, and can be as a method of determining the, content of atractylenoide I and atractylenoide Ⅲ in extracts of the Atractylodis japonica with supercritical fluid extraction.
