摘要
目的:探讨子宫内膜细胞凋亡与诊断超声持续照射时间的关系,以及生殖医学领域应用诊断超声所需安全照射时间。方法:将健康纯种二级Wistar雌性大鼠50只随机分为5组:空白对照组、B超照射10min组、20min组和30min组,彩超照射10min组;照射后24h取子宫内膜,采用流式细胞仪检测子宫内膜细胞早期凋亡的水平。结果:(1)流式细胞仪二维点图显示:对照组未见或偶见散在的早期凋亡细胞;B超照射10min组出现早期凋亡细胞;20min组及30min组的早期凋亡细胞明显增多,以30min组最为明显;彩超照射10min组出现较多的早期凋亡细胞;(2)早期凋亡率比较:照射各组与对照组比较,P<0.01,均有统计学意义,B超照射10min组与20min组及30min组之间均有统计学意义(P<0.01);相同频率彩超照射10min比B超照射10min早期凋亡率明显增高。结论:诊断超声辐照大鼠子宫可诱导内膜细胞凋亡增加,且凋亡率与超声持续时间及输出功率呈正相关。
Objective: To explore the relationships of endometrial apoptosis with continuing exposures of diagnostic ultrasound,and safe exposure of utilization diagnostics ultrasounds in the reproductive medicine.Methods: Fifty Wistar female Rats healthy purebreds second orders were randomLy divided intofive groups: blank group,experiment groups(exposure10min,20min,30min),color Doppler ultrasound exposure 10min.The endometrium were obtained at 24h after exposure,Flow cytometry was used to detect the level prophases apoptosis of endometrial cells.Results:(1)The scatterplot two-dimensions was signify:blank group unemerge or perchance emerge sporadically prophase apoptosis;two-dimension ultrasonic radiation 10min occurrence prophase apoptosis;20min group and 30min group obviously increase,30min group the most obviously;color Doppler ultrasound 10min group appear major prophase apoptosis;(2) The rate ratio prophase apoptosis comparison: experiment groups with contrast,P0.01,two-dimension ultrasonic radiation 10min group and 20min group and 30min group contrast have statistical significance(P0.01);The rate ratio of prophase apoptosis endometriums cells obviously heighten of ultrasonic radiation10min color Doppler ultrasound 10min with two-dimension ultrasonic radiation 10min.Conclusion: Diagnostic ultrasound may result in more uterus cells apoptosis in rats,output power and duration of ultrasound correlates significantly to apoptosis.
