川穹嗪对维拉帕米致急性左心衰竭家兔心功能的影响

Effects of ligustrazine on verapamil-induced acute left heart failure in rabbits

目的:观察不同剂量的川穹嗪对维拉帕米致急性左心衰(AHF)家兔心功能的影响。方法:采用静脉滴注维拉帕米的方法,建立家兔AHF模型。静脉注射不同剂量的川穹嗪观察其对AHF家兔心脏血流动力学的影响。结果:小剂量(8.0 mg/kg)、大剂量(16.0 mg/kg)的川穹嗪均能明显改善AHF家兔的多项血流动力学指标,明显增高AHF家兔的收缩压(SBP)、舒张压(DBP)、脉压(PP)、左室收缩压(LVSP)、左室内压最大上升速度(+dp/dtm ax)以及左室内压最大下降速度(-dp/dtm ax)(n=5,P<0.05),明显降低AHF家兔的左室舒张末压(LVEDP)(n=5,P<0.05)。川穹嗪扩张血管的作用呈浓度依赖性,但其增强心衰心肌收缩力的作用小剂量与大剂量无明显差异。结论:川穹嗪可显著改善AHF家兔的血流动力学参数,有助于AHF的治疗。

AIM： To observe the effects of ligustrazine on cardiac functions of rabbits with acute left heart failure （AHF）. METHODS： Rabbit model with AHF was established by transfusing verapamil through vein. Different doses of ligustrazine were then injected to observe their effects on cardiac hemodynamics of rabbits with AHF. RESULTS： Both low （8.0 mg/kg） and high （16. 0 mg/kg） doses of ligustrazine improved many of the hemodynamic indexes in rabbits with AHF. They significantly increased systolic blood pressure, diastolic blood pressure, pulse pressure, left ventricular systolic pressure, maximum ascending and descending velocity of left ventricular pressure （ ＋ dp/dt - dp/dtmax ） （ n = 5, P 〈 0.05 ）, and decreased left ventricular end-diastolic pressure （LVEDP） （ n = 5, P 〈 0.05）. Ligustrazine dilated vessels in a dose-dependent manner. However, no significantly different effects between low and high doses of ligustrazine on positive inotropic action were found. CONCLUSION： Ligustrazine can markedly improve many hemodynamic indexes in rabbits with AHF and is helpful for the treatment of AHF.
