摘要
目的探讨汽车安全气囊上微量人体脱落上皮细胞DNA检测的有效方法.方法分别采用棉线载体直接扩增法、小体积Chelex-100法和磁珠法对汽车安全气囊上提取的微量人体脱落上皮细胞的DNA进行检测,并对检测结果进行分析比较,以获得有效方法.结果棉线载体直接扩增法和磁珠法均能得到满意的人体脱落上皮细胞核DNA STR基因座分型结果,小体积Chelex-100法不能获得STR基因座分型结果.结论棉线载体直接扩增法和磁珠法均能有效地检测微量人体DNA,获得STR基因座分型结果,提高DNA检出率,操作简单、节约时间和试剂成本,特点在法医物证学实际检案工作中具有较高的应用价值.
Objective To discuss the effective DNA detection method for the trace of human epithelial cells exfoliated on airbag of vehicle.Methods The trace DNA of human epithelial cells extracted form the airbag was detected by the methods of cotton carrier direct amplification,small volume of Chelex-100,and the detection of magnetic beads,and the results were analyzed and compared in order to obtain a valid method.Results Cotton carrier direct amplification method and the detection of magnetic beads method can get the satisfactory STR loci genotyping results of the shedding epithelial cell nuclear DNA of human,whereas the small volume of Chelex-100 method can not obtain the results of STR loci genotyping.Conclusions Cotton carrier direct amplification method and detection of magnetic beads method can detect the trace of human DNA effectively,obtain the results of STR loci genotyping,and improve the DNA detection rate.Especially cotton carrier direct amplification method,which is simple,saving time and reagent costs,and has a high application value in the actual detective work of biological evidence.
