柚皮苷对兔骨髓间充质干细胞分化为成骨细胞的影响 被引量:8

摘要 目的观察柚皮苷(骨碎补的有效成分)对兔骨髓间充质干细胞(BMSCs)分化为成骨细胞的影响。方法将体外培养的兔BMSCs分为对照组(标准培养液)、柚皮苷组(不同浓度柚皮苷+标准培养液)、普通组(地塞米松10-8mol/L+β-甘油磷酸钠10 mmol/L+维生素C 50μg/L+标准培养液)和联合组(不同浓度柚皮苷+普通组)。倒置相差显微镜观察生长情况,MTT法检测不同浓度柚皮苷对兔BMSCs增殖的影响;扫描电镜、茜素红染色法和von Kossa银染色法观察各组细胞钙化情况。结果柚皮苷10-5mmol/L明显促进兔BMSCs增殖。培养3周后,联合组钙结节增多、钙化面积扩大最明显,柚皮苷组可见钙结节;对照组未见钙化。结论柚皮苷可促进兔BMSCs向成骨细胞分化。 Objective To observe the effect of naringin on differentiation of rabbit bone marrow mesenchymal stem cells(BMSCs) into osteoblasts.Methods Rabbit BMSCs cultured in vitro were divided into 4 groups of A(without treatment as the control),B(treated with different concentrations of naringin),C(treated with dexamethasone 10-8mol/L,sodium β-glycerophosphate 10 mmol/L and vitamin C 50 μg/L) and D(treated with dexamethasone 10-8mol/L,sodium β-glycerophosphate 10 mmol/L and vitamin C 50 μg/L plus different concentrations of naringin).The growth state of BMSCs was observed by phasecontrast microscope and the effect of different concentrations of naringin on proliferation of BMSCs was detected by MTT.The calcium node formation was observed by scanning electron microscope,alizarin bordeaux stain and von Kossa stain.Results Naringin 10-5 mmol/L significantly promoted the proliferation of rabbit BMSCs.After 3 weeks,the calcium node formation and calcium area were most apparent in group D.The calcium node formation also occurred in group B,but not in group A.Conclusion Naringin can induce the differentiation of rabbit BMSCs into osteoblasts.
