左旋棉酚对HT1080细胞和HSF细胞的抑制增殖作用 被引量:1

摘要 目的:探讨左旋棉酚对人纤维肉瘤细胞HT1080和人皮肤成纤维细胞HSF的抑制增殖作用。方法:采用MTT法检测药物对细胞增殖的抑制率,在倒置显微镜下观察细胞的形态学变化。结果:左旋棉酚对HT1080细胞和HSF细胞均有抑制增殖作用,呈浓度与时间依赖性。左旋棉酚对HT1080细胞作用24、48、72、96小时的IC_(50)分别为56.6、12.6、9.2、7.0μM,而对HSF细胞作用24、48、72、96小时的IC_(50)分别为192.3、53.6、42.5、19.4μM。随着药物浓度的增加,HT1080细胞数逐渐较少,脱落的细胞逐渐增多。而HSF细胞变化不明显。结论:左旋棉酚对人纤维肉瘤HT1080细胞有明显抑制增殖和选择性杀伤作用。 Objective: To investigate the Effect of (-)-Gossypol on proliferation inhibitory in Cell Line HT1080 and HSE Methods:The proliferation inhibitory rate of cells treated by drugs was deteceted by MTT assay. The changes of cell morphology were detected in inverted microscope.Results:(-)-Gossypol effect on proliferation inhibitory in Cell Line HTI080 and HSF in a time- and dose-dependent manner. The IC^0 value of HT1080 at 24,48,72,96h were 56.6,12.6,9.2,7.0 ~tM and the IC^o value of HSF at 24,48,72,96h were 192.3,53.6,42.5,19.4pM respectively. With the increase of drug concentrations, the number of HT 1080 cells gradually decrease, shedding cells increase gradually. And HSF cells do not change significantly. Conclusions:(-)-Gossypol has significant effect of proliferative inhibition and could selectively kill HT1080 in vitro.
