恶性骨肿瘤中血管内皮生长因子表达的定量研究 被引量:2

Quantitative Study of Vascular Endothelial Growth Factor Expression in Malignant Bone Tumor

摘要 目的观察血管内皮生长因子(vascularendothelialpowthfactor,VEGF)在恶性骨肿瘤中的表达,并探讨VEGF在恶性骨肿瘤中的表达水平与DNA倍体、细胞增殖活性及肿瘤侵袭性的关系。方法应用流式细胞仪及免疫荧光技术检测几种恶性骨肿瘤(骨肉瘤、软骨肉瘤、滑膜肉瘤)的VEGF表达量(荧光指数fluorescenceindex,FI)、DNA含量(DNA指数值DNAindex,DI)及细胞增殖活性(增殖指数prolifewtionindex,PI)。结果在恶性骨肿瘤中多数为DNA异倍体,各类恶性骨肿瘤之间FI、DI、PI值差异无显著性意义(P>0.05)。包膜完整组与包膜不完整或无明显包膜组之间FI值差异有非常显著性意义(P<0.01);异倍体与二倍体肿瘤之间n值差异有非常显著性意义(P<0.01);VEGF表达阳性组与VEGF表达阴性组之间PI值差异有非常显著性意义(P<0.01);直径>5cm组与直径≤5cm组之间FI值差异无显著性意义(P>0.05)。结论VEGF表达水平与恶性骨肿瘤的侵袭性、DNA倍体及细胞增殖活性密切相关,而与肿瘤大小无明显相关。提示VEGF可能作为判断骨肿瘤恶性程? Objective To observe vascular endothelial growth factor (VEGF) expression in malignantbone tumor and the relationship between VEGF expression and DNA ploidy, cellular proliferation activity, in-vasion of malignant bone tumor. Methods The expression of VEGF (nuorescence index, FI), DNA content(DNA index, DI), cellular proliferation activity (proliferation index, PI ) in malignant bone tumor (osteosarcoma,chondrosarcoma, synovial sarcoma) were detected using now cytometry and immunofluorescence staining techniqUe. Results Majority of malignant bone tumor are aneuploid. There isn' t significant difference in FI, DI, PIof each type of malignant bone tumor (P > 0. 05 ). There is great significant difference in FI between the intactcapsule group and unintact capsule group (P < 0. 01 ), in FI between DNA aneuploid group and DNA diploidgroup(P < 0.01 ) and in PI between VEGF expression positive group and VEGF expression negative group (P <0. 01 ). There isn' t significant difference in FI between diameter > 5 cm group and diameter≤5 cm group (P >0. 05). Conclusion The expression of VEGF is highly related to the invasion, DNA ploidy and cellular proliferation activity of malignant bone tumor. It isn't related to the size of tumor. The expression of VEGF is probablya predictor for judgment of malignant degree of bone tumor.
