用荧光检测方法研究氦-氖激光对小鼠腹腔巨噬细胞的活化作用 被引量:1

摘要 目的:探讨氦- 氖(He- Ne) 激光对机体的免疫刺激作用的机理。方法:用剂量为63-7J/cm2 的He-Ne 激光照射小鼠的神阙穴,时间分别为3 ,5 ,7 和10 d ,每天照射5 min,然后分别做如下实验:a.用荧光显微技术观察腹腔巨噬细胞( Mψ) 对白色念珠菌的吞噬功能;b .α- 萘酚醋酸酯法显示腹腔Mψ内非特异性酯酶(NSE),并用图象分析系统对NSE作定量分析。结果:各照射组吞噬率、吞噬指数及NSE 的各项参数值均较对照组有非常显著性差异( P< 0-01) ,随照射天数增加,各项指标也随之增加,各照射组间比较也均有非常显著性差异( P<0-01)。结论:适当剂量的He- Ne 激光能活化Mψ,提高机体免疫功能。 Objective:To research He-Ne laser stimulating immunity of organism. Methods:Shenque acupoint of mice abdomen was irradiated by He-Ne laser at the dosages of 63.7J/cm 2 for 3,5,7 and 10 days respectively, 5 minutes each day. Then following methods are taken:a.Observing the phagocytosis to C. albicans of mice peritoneal macrophages (PMψ) with the fluorescence microscope after acridine orange (AO) staining. b.Measuring the non-specific esterase(NSE) in PMψ after being stained with α-naphthyl acetate by image analysis. Results:The above indices of irradiated groups were higher than that of non-irradiated group (P<0 01). Comparison between different irradiated groups also showed significant difference(P<0 01). Conclusion:It is suggested that the He-Ne laser with appropriated dosages can activate immunocytes, and enhance the immunity of organism.
