摘要
目的:探讨巨核细胞集落生成单位(CFU- Meg) 对辐射的敏感性。方法:(C57BL/6J)小鼠经60Co γ射线全身一次均匀照射,剂量为3Gy 。取骨髓细胞体外培养9 d,乙酰胆碱酯酶染色,吉姆萨染色,计数,观察。结果:照射后2 ~3 d 巨核细胞数量明显下降,小乙酰胆碱酯酶阳性细胞基本降至零;第5 d 数量回升后再次下降,7 d 后巨核细胞的数量缓慢上升,但仍低于对照组。结论:CFU- Meg 对辐射具有较高的敏感性;较成熟的巨核细胞对辐射有较强的抗性。
Objective:To study the radiosensitivity of colony-forming unit-megakaryocyts (CFU-Meg). Methods:After dose of 3Gy 60 Co γ-rays total body irradiation, C 57 BL/6J mouse bone marrow cells were incubated in culture for up to 9 days, stained by acetylcholinesterase and Giemsa. The megakaryocytes(Meg) were observed morphologically and the number of Meg were counted. Results: The number of Meg decreased markedly during the first 3 days after irradiation, SAchE(+) declined nearly to zero. The number of Meg increased slightly at 5th day and then reduced again. The number of Meg started to increase gradually after 9th day but were still lower than that in control mouse. Conclusion: Based on the obtained results, CFU-Meg showed higher degree of sensitivity to γ-rays and more mature Meg showed higher degree of resistance to radiation injury.
