摘要 目的探讨多氯联苯对P19细胞向心肌分化进程的影响。方法在P19细胞向心肌细胞分化的培养液中加入多氯联苯(Aroclor 1254)2.5μmol/L,同时设立阴性对照组。显微镜下连续观察P19细胞形态的变化,并采用实时荧光定量RT-PCR技术对分化过程的第0、4、10天中GATA4及Nkx2.5基因mRNA的表达水平进行检测。结果在分化全程(0~10天),实验组与对照组无细胞形态上的差别;而GATA4、Nkx2.5基因的表达量除在分化开始前(第0天)差异无统计学意义外,在分化中期(第4天)及末期(第10天),实验组中GATA4、Nkx2.5基因的相对表达量均显著低于对照组。结论多氯联苯可通过对早期心脏发育相关重要基因的表达影响心肌分化进程。 Objective To explore the effect of polychlorinated biphenyls(PCBs)during the differentiation of P19 cells to cardiac myocytes.Methods P19 cells were treated with 2.5 μmol/L PCBs(Aroclor 1254)and untreated cells were used as control.The morphologic changes of P19 cells were examined and photographed using an inverted microscope.The expressions of GATA4 and Nkx2.5 mRNA were evaluated by real-time quantitative RT-PCR on days 0,4,and 10 of differentiation.Results There was no significant difference in morphology between P19 cells treated with PCBs and controls.On day 0,the expression of GATA4 and Nkx2.5 mRNA was not significantly different between P19 cells treated with PCBs and controls.However,on day 4(the middle of differentiation)and 10(the end of differen-tiation),the expression of GATA4 and Nkx2.5 mRNA were significantly lower in P19 cells treated with PCBs than controls.Conclusions The PCBs affected differentiation of cardiac myocytes by regulating genes related to heart development.
