丹参酮IIA磺酸钠对兔耳增生性瘢痕中MMP-1mRNA表达的影响

Effect of Sodium Tanshinone IIA Sulfonate on MMP-1mRNA expression for hypertrophic scars of rabbit ears

目的通过基因检测和组织染色的方法,来研究丹参酮IIA磺酸钠对兔耳增生性瘢痕中MMP-1mRNA表达的影响和其对增生性瘢痕的作用。方法①先建立兔耳增生性瘢痕模型,以曲安奈德为阳性对照,分组分次瘢痕基底局部给药,取下瘢痕组织,使用RT-PCR方法检测细胞内金属基质蛋白酶MMP-1基因表达;用苏木素-伊红染色(HE)、苦味酸酸性复红染色法(VG)染色法对成纤维细胞和胶原纤维进行观察。结果 RT-PCR基因检测显示丹参酮及曲安奈德组的MMP-1mRNA的表达显著高于空白对照组;HE染色显示空白对照组中成纤维细胞数量明显高于丹参酮IIA磺酸钠组,丹参酮IIA磺酸钠组与曲安奈德组无明显差异;VG染色显示丹参酮IIA磺酸钠组胶原纤维排列较空白对照组显著整齐,与曲安奈德组无明显差异。结论丹参酮IIA磺酸钠能够通过增强MMP-1mRNA的表达及抑制成纤维细胞的增殖,减少胶原纤维的合成而抑制瘢痕的过度增生。

Objective Through genetic testing and tissue staining method,to research effects of sodium tanshinone IIA sulfonate on MMP-1mRNA expression for hypertrophic scars of rabbit ears and hypertrophic scars.Methods First rabbit ears were used to establish animal models for hypertrophic scars,triamcinolone acetonide as a positive control,the hypertrophic scars were injected intralesionally with medicines or placebo for different groups and different times.Then remove the scars,detected matrix metalloproteinase MMP-1 gene expression by using RT-PCR analysis;observed fibroblasts and collagen fibers by using hematoxylin-eosin staining（HE） and Van.Gieson（VG） staining.Results RT-PCR gene analysis showed that MMP-1mRNA expressions for tanshinone and triamcinolone acetonide were significantly higher than placebo;HE staining showed the number of fibroblasts of placebo was significantly higher than sodium tanshinone IIA sulfonate group,there was no significant difference between sodium tanshinone IIA sulfonate group and triamcinolone acetonide group;VG staining showed that collagen fibers for sodium tanshinone IIA sulfonate group arranged significantly more neatly than for placebo group,but it was no significant difference between triamcinolone acetonide group.Conclusion It is suggested that sodium tanshinone IIA sulfonate could inhibited the excessive proliferation of scars through enhanced expression of MMP-1mRNA and inhibition of fibroblast proliferation and reduced the synthesis of collagen fibers.
