摘要
目的:了解大连地区人群NA基因频率及NA基因的多态性。方法:分别从大连地区111 名随机健康献血者抽取500μl(EDTA抗凝)全血,经酚—氯仿法抽提DNA,采用聚合酶链反应-序列特异性引物(PCR-SSP)技术作NA系统的DNA 分型。从基因组DNA直接扩增、电泳(溴乙锭染色液加入琼脂糖中)后,紫外灯下判读结果,指定等位基因特异性。结果:大连地区人群NA1 基因频率为0.446,NA2 基因频率为0.554。结论:大连地区人群中NA2 基因较多,但与NA1
Objective:To understand the neutrophil antigen(NA) gene frequencies and the NA gene polymorphism in the Dalian region population.Methods:The genomic DNA was extracted by phenol chloroform extraction method from 500μl of EDTA anticoagulant whole blood which had been drawn from 111 randomly recrutied healthy blood donors without blood relationship in Dalian region,respectively.The DNA of NA gene system was typed by polimerase chain reaction with sequence specific primers(PCR SSP).After the direct amplification and electrophoresis(the bromoacetic troche staining solution added to agarose) from genomic DNA,the result reading was carried out by UV,and then the allete specificity was assigned.Results:The frequency of NA1 gene and NA2 gene in the Dalian region population was 0 44 and 0 554,respectively.Conclusion:There are more NA2 genes in the Dalian region population,but there is a little difference between the NA1 gene frequency and the NA2.
