摘要
为建立更加安全高效的辣椒多倍体诱导技术体系,采用辣椒兴蔬201子叶离体组织培养技术,将分化出的丛生芽进行抗微管除草剂氟乐灵的诱导试验。结果表明:供试材料均出现明显的根尖加粗现象,利用根尖压片和保卫细胞气孔面积进行分析,添加50 mg/L氟乐灵,处理时间为24 h获得的加倍效果最好,加倍诱导率达26.5%,比对照秋水仙碱处理的最高加倍诱导率(23.2%)提高了3.3%,植株平均致死率下降了12.2%。表明,利用氟乐灵处理辣椒离体组织是获得四倍体辣椒的有效途径之一。
Cluster buds differentiated from the in vitro tissue culture of cotyledon of hot pepper variety Xingshu 201 were induced by anti-microtubule herbicides trifluralin to develop a safe and efficient polyploid induction technology system for hot pepper.The results showed that all tested materials appeared root tip emboldening.According to the observation of root tip squashing slides and areas analysis of guard cells and stomata,the treatment with 50 mg/L of trifluralin and 24 hrs of treatment time had effective chromosome doubling with double induction rate of 26.5%.The induction rate increased by 3.3% compared with the control treated by colchicines with rate of 23.2%,and the plant mortality decreased by 12.2% in this treatment.It indicated that it was one of the most effective solutions for tetraploid hot pepper induction to treat the in vitro hot pepper tissues using trifluralin.
