低分子RNA及5-Fu对白血病细胞系K562细胞抑制作用的实验观察

摘要 目的:观察低分子RNA 及5- Fu 在白血病细胞系K562 细胞中的作用。方法:在K562细胞培养中加低分子RNA( 实验组1) 、5 - Fu( 实验组2) 、低分子RNA 和5 - Fu( 实验组3) 及P-B-S( 对照组),观察集落细胞数,死、活细胞计数及细胞蛋白含量。结果:集落细胞数对照组为142 个,实验组1 为70 个,抑制率为50-7 % ,实验组2 为71 个,抑制率为50-5% ,实验组3 为41个,抑制率为71-1 % ;活、死细胞实验组1 为42 个、11 个,实验组2 为43 、5 个,实验组3 为27 个、16 个;细胞蛋白含量实验组1 为1-610 ,实验组2 为1-622 ,实验组3 为0-875 ,占72-7 % 。结论:低分子RNA能对白血病细胞系K562 细胞的增殖有明显抑制作用,且能增强5 - Fu 抑制肿瘤细胞的增殖效果。 OBJECTIVE:To evaluate effect of low-molecular RNA and 5-Fu inhibiting cell proliferation of leukemic K562 cells. METHOD:Add low-molecular RNA(group one),5-Fu (group two),low-molecular RNA and 5-Fu (group three) and P.B.S (control group) to culture medium of K562 cells.Cell clusters live and dead cells were counted and cell protein was detected.RESULTS:Number of cell clusters:70 in group one,inhibiting rate 50.7%;71 in group two, inhibiting rate 50.5%;41 in group three,inhibiting rate 71.1%.Number of live and dead cells:42 and 11 in group one, 43 and 5 in group two, 27 and 16 in group three. 43 and 5 in group two,27 and 16 in group three.Cell protein:group one 1.610,group two 1.622 and group three 0.875, making up 72.7%.CONCLUSION:Low-molecular RNA can remarkably inhibit proliferation of leukemic K562 cells and strengthen the effect of 5-Fu inhibiting Proliferation of tumor cells.
