Effects of hyperbaric oxygen on the apoptotic splenocytes in normal rats and rats with systematic inflammatory response syndrome

摘要 目的实验观察高压氧(HBO)对于正常机体及系统性炎症反应综合征(SIRS)状态下脾细胞凋亡的影响。方法 健康雄性SD大鼠50只,体重140~180g,随机分为5组,每组10只。A组:腹腔注射等量生理盐水;B组:腹腔注射等量生理盐水,做3次HBO;C组:腹腔注射酵母多糖-石蜡悬液(500mg/kg);D组:腹腔注射酵母多糖-石蜡悬液(500mg/kg),做1次HBO;E组:腹腔注射酵母多糖-石蜡悬液(500mg/kg),做3次HBO(2ATA)。24小时后杀死大鼠,取脾,用凋亡检测试剂盒标记凋亡脾细胞,用流式细胞仪检测脾细胞凋亡比例。结果HBO治疗对正常大鼠脾细胞凋亡无明显影响,与正常对照组比较,P(0.05,SIRS状态下大鼠脾细胞凋亡明显增加,与正常大鼠组比较,P〈0.05,HBO明显减少SIRS组大鼠脾细胞凋亡率,与SIRS组比较,P〈0.05。结论 HBO根据机体处于不同状态而对脾脏细胞凋亡产生不同的影响。 Objective To investigate the effect of hyperbaric oxygen (HBO) therapy on the apoptetic splenocytes in normal rats and rats with systematic inflammatory response syndrome (SIRS). Methods SD rats were randomly divided into five groups, with ten rats for each group. Group A were treated by saline solution (5mL/kg) ,i. p. ; Group B were treated by saline solution and three HBO (2ATA) treatments; Group C were treated by zymosan (500mg/kg) ,i. p. ; Group D were treated by zymosan and a single HBO exposure; Group E were treated by zymosan and three HBO exposures. All animals were killed 24 hours after intraperitoncal injection and spleen samples were prepared for analysis of apoptotic splcnocytes by flow eytometer. Results HBO has no effect on apoptotic splenecytes in normal rats, without significant difference compared to those in normal group ( P 〉 0.05), the percentage of apoptotic splenocytes in SIRS group was significantly increased compared to that in normal group ( P 〈 0.05), HBO significantly decreased the percentage of apoptetic splenocytes in rats with SIRS compared to those in SIRS group ( P 〈 0.05). Conclusion HBO may play different effects on apoptotic splenocytes depending on different status of the host.
