小鼠胡须体觉皮质局灶性脑缺血模型建立 被引量:2

Construction of Whisker Barrel Cortex Focal Ischemic Animal Model in Mouse

摘要 目的:制作梗死区集中在小鼠大脑胡须体觉皮质的局灶性脑缺血模型。方法:小鼠麻醉后,经右侧顶叶皮质处开颅,显微镜和内在光信号成像下结扎大脑中动脉分支2~3支。应用激光多普勒血流仪检测结扎前、后局部脑血流情况,TTC染色检测梗死面积大小,TUNEL染色标记细胞死亡情况,细胞色素氧化酶染色确定体觉皮质和梗死部位的关系。结果:大脑中动脉分支结扎后,梗死中心及周围局部脑血流明显下降。缺血后3 d,TTC染色示梗死区位于胡须体觉皮质,梗死面积约占脑冠状切面面积的15%,TUNEL染色示死亡细胞位于胡须体觉皮质,不累及白质,细胞色素氧化酶染色示胡须体觉皮质结构破坏。结论:可选择性将小鼠具有单一功能的胡须体觉皮质制作成局灶性脑缺血模型,为脑缺血研究的动物模型提供更多选择。 Aim:To set up a focal cerebral ischemia animal model that the infarct region targets at whisker barrel cortex of mice.Methods:The mice were subjected to anesthesia,and the 2~3 branches of right middle cerebral artery(MCA) supplying the barrel cortex were permanently ligated under the direction of microscope and intrinsic optical signal imaging.Before and after ligation,local cerebral blood flow was assessed by laser Doppler scanner.The infarct volume was measured by TTC staining.Cell death was assessed by TUNEL staining.Barrel pattern on the cortex was examined using cytochrome oxidase staining.Results:The local cerebral blood flow significantly decreased after the branches of MCA were ligated.TTC staining revealed that 3 days after stroke the infarct region which targeted at the barrel cortex was about 15% of the whole brain.Cell death revealed by TUNEL staining focused on the barrel cortex without damaging the white matter.Cytochrome oxidase staining revealed the ischemic damage occured in the barrel cortex.Conclusion:It is possible to set up a focal cerebral ischemia animal model that the infarct region target at whisker barrel cortex of mice,which provides more alternatives when selecting animal model for the studies of cerebral ischemia.
