摘要 目的:通过大鼠游泳方式建立过度运动肾脏损伤模型。方法:大鼠随机分为对照组和力竭游泳组。力竭组大鼠在玻璃钢泳池中游泳至力竭。于力竭后不同时相(0、6、12、24h)取材,检验肾功能及尿γ-GT浓度,通过光镜及电镜观察大鼠肾脏病理变化,并采用TUNEL法检测肾脏组织细胞凋亡。结果:游泳至力竭后大鼠表现极其疲劳,尿素氮开始升高,力竭后6h肌酐也上升。力竭后大鼠肾脏光镜及电镜观察均有损伤性改变,且随时间延长损伤有加重趋势。TUNEL结果显示力竭后6h肾组织细胞凋亡率较对照增高明显(P<0.05)。结论:游泳至力竭后,大鼠肾脏无论功能还是结构都发生损伤性变化,大鼠单次游泳至力竭模型可以做为研究过度训练致肾脏损伤的理想模型。 Objective:To establish the rat kidney injury model by exhausting swimming.Methods:Forty rats were randomly divided into two groups :control group(CN,n=8)and exhausting swimming group(ES).The rats of ES swim in pool to exhaustion.We had collected the specimen in different phase after exhausting swimming.We assayed the kidney function and the urine γ-GT.The renal pathological change was observed through the light microscope and the electron microscope.The apoptosis of kidney was detected by TUNEL stain.Results:The performance of rats in ES group was extremely weary after exhausting swimming.The urea nitrogen started to elevate immediately,then the creatinine elevated too.The rats' kidney had been damaged after exhausting swimming observed by light microscope and the electron microscope.The damage change had the aggravation tendency along with the time.The cell numbers of apoptosis also increased gradually after exhausting swimming acrossing the TUNEL result(P0.05).Conclusion:The rat kidney has change including function and pathological after the exhausting swimming.Letting rat swim to exhaustion is a good model to reseach the renal injury by overexercise.
