雷公藤甲素增强5-FU对肝癌细胞SMMC 7721诱导凋亡作用的实验研究

摘要 目的观察雷公藤甲素(triptolide,TPL)5、-FU对肝癌细胞SMMC 7721增殖及凋亡的影响;探讨TPL能否增强5-FU对SMMC 7721细胞的诱导凋亡作用。方法 SMMC 7721细胞置于RPMI-1640培养液+10%胎牛血清中培养;MTT法测定5-FU、TPL对SMMC 7721细胞的增殖抑制效果;Western Bloting方法检测凋亡相关蛋白PARP的表达变化;流式细胞仪方法分析细胞凋亡比例的差别。结果 TPL、5-FU对AsPC-1细胞均具有抑制增殖作用,其48 h IC 50分别为:11.14 ng/mL±1.31 ng/mL和2.34μg/mL±0.39μg/mL。TPL与5-FU联合应用于SMMC 7721细胞,联合用药组与各单药组比较,细胞凋亡比例及cleaved PARP表达量均明显增加,差别均有明显统计学意义(P<0.01)。结论 TPL可明显增强5-FU对肝癌细胞SMMC 7721的诱导凋亡效果。 Objective To observe the effects of triptolide(TPL) and 5-fluorouracil on hepatocellular carcinoma(SMMC-7721) cells growth and apoptosis.To investigat whether TPL could enhance 5-fluorouracil-induced apoptosis in SMMC-7721 cells.Methods SMMC-7721 cells were cultured in the culture medium with RPMI-1640 and 10% fetal calf serum.Inhibited proliferation effect of 5-fluorouracil and TPL on SMMC-7721 cells were determined by MTT assay.The expression changes of PARP were detected by Western Bloting.The cell apoptosis ratios were analyzed by flow cytometer.Results The proliferation of SMMC-7721 cells could be inhibited by TPL and 5-fluorouracil.Their IC50 on 48 h were 11.14±1.31 ng/mL and 2.34±0.39 μg/mL respectively.TPL combined with 5-fluorouracil were used on SMMC-7721 cells.The expression amount of cleaved PARP and the apoptosis proportion in combined drug group were increased obviously than in each single drug group,and both results have statistically significant differences(P0.01).Conclusion TPL could obviously enhance 5-fluorouracil-induced apoptosis in hepatocellular carcinoma(SMMC-7721) cells.
