摘要 采用吐温60强化酶法-微波提取墨旱莲总黄酮,选取纤维素酶质量浓度、酶解温度、酶解时间、料液比、吐温60质量浓度、微波提取时间进行六因子五水平二次正交旋转组合试验,确定提取的最优条件,将优化后的提取结果与其他提取方法进行比较。结果表明:优化的提取条件为纤维素酶质量浓度1.00 g/L、酶解温度54℃、酶解时间90 min、料液比21 mL/g、吐温60质量浓度2.4 g/L、微波提取时间142 s;在最优条件下测得墨旱莲黄酮得率为1.38%。与其他提取方法相比,吐温60强化酶法-微波节能高效。 The extractive process of combining tween 60 with enzyme and microwave was used to extract total flavones from Eclipta alba.The quadratic orthogonal rotation design with six factors and five levels was designed to obtain the optimum extraction parameters.Extraction condition were as follows: the cellulase concentration of 1.00 g/L,enzymatic temperature of 54 ℃,enzymatic time of 90 min,liquid to solid ratio of 21 mL/g,tween 60 concentration of 2.4 g/L,and microwave extraction time of 142 s.Under the conditions,the yield of the total flavones was 1.38%.Compared with other methods,the extractive methods by combining tween 60 with enzyme and microwave was a energy-saving and high-efficiency one.
