摘要
AFLP是以PCR扩增为基础的,是扩增片段长度多态性的简称。它是RFLP和PCR相结合的一种标记技术,其基本原理是对基因组DNA限制性酶切片断的选择性扩增。AFLP的基本反应过程包括模板DNA制备、酶切片段的扩增和扩增产物的凝胶电泳分析3个步聚。AFLP是一种新颖和强大的DNA指纹技术,结合了RFLP和RAPD的优点,具有高效、快捷、稳定、可靠的特点。本文将AFLP和RFLP、RAPD、SSR等分子标记进行了比较,结果表明它可产生丰富而稳定的遗传标记,因而在林木分类、品种鉴定、遗传图谱构建、育种和目的基因定位等研究上有良好的应用前景。
Based on PCR,AFLP is the abbreviation of amplified fragment length polymophism.Its a technique of marker which is connected with RFLP and PCR.Its principle involves the selective amplification of a genomic DNA digests using PCR.The method involves three basic steps:(1)template preparation(2)selective amplification(3)denaturing polyacrylamide gels analysis of AFLP products following by autoradiography or non-autoradiography.AFLP is a novel and strong fingerprint technique of DNA.It is connected with the virtue of RFLP and RAPD,so it has characteristics:high efficiency,promptitude,stability,accuracy.The article compares AFLP with RFLP,RAPD,SSR.The result demonstrate:AFLP can produce abundant and stable genetic marker,so it has better prospect on classification,variety identification,genetic mapping,breeding and location of target gene in the forestry.
