刺参酸性粘多糖的分离及鉴定 被引量:11

摘要 将刺参体壁用0 .5 molLK2CO3 抽提,胰酶水解,乙醇沉淀得多糖粗品,再经过氧化氢脱色、萃取脱蛋白质、透析除盐等工艺制得多糖纯品.此多糖纯品经凝胶电泳和醋纤薄膜电泳证明为均一性多糖.红外光谱分析表明,样品具有典型的多糖吸收峰.薄层色谱分析确定其单糖组成为葡萄糖醛酸、氨基半乳糖和岩藻糖.此外,样品富含硫酸基团,属典型的酸性杂多糖,HPLC测定其平均相对分子量为50 000 . After extraction with K 2CO 3 and hydrolysis with enzyme,the body wall of Stichopus japonicus was fractionally precipitated with alcohol,then decolourized with H 2O 2 and further purified by Sevag method,the acidic polysaccharide(APS) was obtained.The APS is shown to be pure by agarose gel electrophoresis and cellulose acetate film electrophoresis.The analysis of IR spectrum scanning shows a characteristic absorption bands of polysaccharide.Its monosaccharide composition is identified by thin layer chromatography.The results indicate that the APS consists of D galactose,rhamnose galacturonic acid and abundant sulfate.Its average relative molecular weight is estimated to be 50 000 by HPLC.
