ROCK信号通路在流体剪切力诱导的成骨细胞的细胞骨架改建中的作用研究 被引量:6

摘要 目的:探讨ROCK信号在流体剪切力介导的成骨细胞的细胞骨架改建中的作用。方法:使用平行平板流室系统对体外培养的小鼠原代成骨细胞加载12dyn/cm2的流体剪切力1h,实验组的细胞在加载前使用ROCK的特异性抑制剂Y-27632(10μmol/L)预处理细胞1h。用激光共聚焦显微镜观察并比较Y-27632对流体剪切力介导的小鼠原代成骨细胞肌动蛋白细胞骨架重构的影响。结果:成骨细胞经流体剪切力加载后,肌动蛋白细胞骨架聚合增强,应力纤维的排列趋向一致;事先使用Y-27632预处理细胞并进行加载处理,可抑制流体剪切力所致的成骨细胞内肌动蛋白细胞骨架聚合和重建。结论:RhoA-ROCK信号通路调控着流体剪切力介导的成骨细胞细胞骨架改建。
