非创伤性下肢缺血预处理琥珀酸脱氢酶电镜细胞化学的变化

摘要 目的:通过琥珀酸脱氢酶(SDH) 电镜细胞化学的变化,探讨非创伤性下肢缺血预处理的心肌保护效应。方法:实验采用体重250 ±30g ,雄性SD 大鼠24 只,分4 组,每组6 只,即正常对照组( Ⅰ组) ;缺血再灌注组( Ⅱ组) ;经典缺血预处理组( Ⅲ组) ;非创伤性下肢缺血预处理组( Ⅳ组) 。按Nitro - BT 法显色制作SDH 光镜标本,按小川的亚铁氰化铜法制作电镜标本。光镜观察SDH 活性参照Niles 法分为4级。结果:Ⅰ组SDH 活性和心肌结构相对正常;Ⅱ组SDH 活性降低明显,线粒体损伤严重;Ⅳ组近于Ⅰ组;Ⅲ组界于Ⅰ组、Ⅳ组之间。结论:非创伤性下肢缺血预处理具有与经典缺血预处理相类似的抗心肌缺血再灌注损伤的效应。 Objective:This aims of this study were exployed the effect of the non-wounded legs repeated-brief ischemic preconditioning by means electron microscopy of succinic dehydrogenase (SDH)to futher provide some experimental data for non-injury IPC,method.Methods:24 male SD rats ,Weighting 250±30g,were divided into 4 groups,each of which consists of 6 rats ,they are normal control group(NC);ischemic-reperfusion group (I/R);classical ischemic preconditioning group(IPC)and non-wounded legs ischemic preconditioning(N-WIPC),After finishing the operation,the myocardium of left ventricle was frozen sectioned and demonstrated SDH activity with Nitro-BT method. The clectron microscopic samples for SDH activity were prepared according to ferrocyanide copper of Ogawa.The activity of SDH was divided into 4 scales based on Niles classification.Results:The result indicated that as compared with NC group the injury on mitochondria and the decrease on SDH activity were the most serious in I/R group ,but is diminished greatly in N-WIPC group. The changes of IPC group is between the two groups above.Conclusion:The study suggested that the non-wounded legs ischemic preconditioning showed the anti-injury in myocardium Caused by ischemic /reper fusion,which is similar to the effect of classical ischemic preconditioning.
