铝对体外培养人胚成骨细胞毒作用的形态学观察 被引量:3

摘要 目的 :观察不同剂量的铝对体外培养的人胚成骨细胞毒作用。方法 :采用无血清体外细胞培养方法 ,将成骨细胞分成对照组、低铝组和高铝组 ,以光镜和电镜观察其形态学变化。结果 :光镜观察 ,高铝组细胞存活数明显低于对照组和低铝组 ( P<0 .0 5) ,高铝组细胞变性坏死率明显高于对照组和低铝组 ( P<0 .0 1 ) ;电镜观察 ,高铝组细胞核膜迂曲肿胀 ,线粒体结构不清晰 ,胞质内含有大量的次级和终末溶酶体 ,多数为髓鞘样结构 ,而低铝组变化轻微。结论 :高剂量铝可抑制体外培养的人胚成骨细胞生长 ,对成骨细胞具有一定的毒性作用。 Objective:To study the toxic effect of aluminum on human embryonic osteoblasts in vitro Methods:The human embryonic osteoblasts were divided into three groups (control, low and high Al groups) The growth, development and morphology of osteoblasts were observed by the light and electron microscope Results: The observation of light microscopy showed that the survival cell quantity in high Al group was remarkably lower than those in the control and low Al groups ( P <0 05), and the detective rate of cell degeneration and necrosis was more than those in the two groups ( P <0 01) The observation of electron microscopy showed that the nucleic membranes in high Al group were tortuous and swollen slightly, the bilayer structure of nucleic and mitochondia membranes was not clear There were a large number of secondary and ultimate lyosomes in high Al group and the majority of them were myelinlike structures But the changes of low Al group wre slight Conclusion: The growth and development of human embryonic osteoblasts was inhibited in high Al group The effect of high dose Al on the ultrastrucure of osteoblasts mainly showed the damage of the membrane structure of cells
