摘要
目的:了解四氧嘧啶致小鼠糖尿病模型建立的实验条件和主要影响因素。方法:观察小鼠性别、体重、禁食时间、四氧嘧啶剂量、四氧嘧啶溶媒的不同组合,对造模成功率、死亡率的影响。结果:选择体重27~29g的雄性小鼠禁食24 h,并用pH值为4.5柠檬酸缓冲液溶解的160 mg/kg四氧嘧啶腹腔注射,获得成模率较高、死亡率较低、血糖较持续稳定的糖尿病小鼠模型。结论:本次造模条件组合优化,充分考虑了各影响因素之间的关联与制约,筛选出较理想的造模条件。
Objective:To optimize fit together the experimental conditions for establishing a diabetes mice model with alloxan.Methods: The experimental influential factors including solution,fasting time,dose and mode delivered by alloxan have been fitted together optimization to evaluate model successful rates and death rates and,opting for mice gender and weight ranges.Results: A better diabetes mice model which with higher successful fitting rate and lower death rate and keeping sustainable stable blood sugar level,can be gained by choosing 27-29g male mice with abdomen injection of 160mg/kg alloxan in pH4.5 citric acid buffer after being fasted for 24 hour.Conclusion: It has fully considered the inter-connections and constraints in this optimization of fitting together the experimental conditions,so as to screen more idea fitting together the experimental conditions.
