摘要 目的探讨二烯丙基二硫(DADS)对人白血病K562细胞增殖、诱导凋亡和细胞周期的影响。方法采用CCK-8技术检测细胞存活率,进行细胞形态学观察,DNA琼脂糖凝胶电泳和流式细胞仪检测DADS对细胞凋亡及周期的影响。结果 DADS能有效地抑制人白血病K562细胞增殖,15、30、60、120和240μmol/L DADS作用于K562细胞48h后,抑制率分别为26.3%、58.2%、44.7%、62.6%和75.6%。Hoechst形态学观察呈现典型的凋亡形态学改变,琼脂糖凝胶电泳上呈现出凋亡特征性的梯状条带,流式细胞仪检测可见亚二倍体峰并阻滞细胞于G2/M期,30、60和120μmol/L DADS作用于K562细胞12h后,Annexin V/PI双标记法检测早期凋亡细胞率分别为18.59%±0.81%、20.85%±0.65%、23.5%±1.1%(P<0.05)。结论 DADS呈浓度依赖性抑制K562细胞生长,其机制与诱导细胞凋亡并阻滞细胞于G2/M期有关。 Objective Diallyl Disulfide(DADS) is a natural compound derived from crushed garlic.This paper is to explore the proliferation inhibited,apoptosis induced and cell cycle regulated effect of DADS on human K562 cells.Methods The CCK-8 assay,morphological analysis,DNA fragmentation technique and flow cytometry analysis were used.Results After 48 hours of treatment with(15,30,60,120,240 μmol/L) DADS,the inhibitory rate of human K562 cells was 26.3%,58.2%,44.7%,62.6%,75.6%(P〈0.05) ,respectively.Meanwhile the K562 cells showed typical apoptosis changes,a typical sub-peak before G0/G1 was observed by flow cytometry and cell cycle was arrested in the G2/M phase,after 12 hours treatment with DADS at the different concentration(30,60,120 μmol/L) ,the rate of apoptosis of K562 cells was 18.59%±0.81%,20.85%±0.65%,23.5%±1.1%,respectively.Conclusion The DADS-treated K562 cells showed a dose-dependent decrease in cell viability and proliferation,and its possible mechanism was involved in apoptosis and G2/M arrest induced by DADS.
