摘要
采用717强碱性阴离子交换树脂,在对其进行酸碱处理后,利用得到的Cl-型交换树脂来固定黑豆酯酶。通过设计单因素和正交优化试验,得到黑豆酯酶的较佳固定化条件。结果表明较佳的固定化条件为:粗酶液稀释50倍,固定化温度为25℃,固定化时间为9 h。此条件下固定化酶回收率为55.87%。对氧乐果的抑制率试验表明:此固定化黑豆酯酶对氧乐果的抑制程度与氧乐果的浓度变化有良好的线性关系,对应的回归方程为y=17.605x+57.437。
Black soybean esterase was immobilized by Cl-type of 717 anion exchange resin coped with acid and alkali solution.Then optimun immobilized conditions were obtained through single factor and orthogonal test.Results show that optimun immobilization conditions were a diluted 50 times solution of crude enzyme,an immobilization temperature of 25 ℃,and an immobilization time of 9 hours.Under these conditions.Meanwhile,the recovery rate of immobilized enzyme was 55.87 %.The inhibition experiments to the Omethoate showed that: the immobilized enzyme had a good linear relationship with the concentration change of Omethoate.The corresponding regression equation was y=17.605x+57.437
