摘要
文章以北陆蓝莓叶片为材料,研究酶液配比浓度、酶解时间、温度、pH值以及甘露醇浓度对蓝莓叶片原生质体制备的影响。结果显示,以2.0%纤维素酶+0.8%果胶酶为酶液组合,0.60 mol/L甘露醇为渗透压稳定剂,酶液pH值为5.8,温度(26±1)℃,黑暗条件下酶解8 h,此后以1 000 r/min,离心6 min,所得蓝莓叶片原生质体的产量与活力最优。试验材料由辽东学院农学院蓝莓养殖基地提供。
The effect of enzyme solution proportion and density,enxymolysis time,temperature,pH value and manitol density on protoplast preparation from leaves of northland blueberry was studied.The results showed that the optimal enzyme solution for protoplast isolation was 2.0% cellulose added with 0.8% pectolgase with 0.60 mol/L mannitol as the osmosis stabilizer.The pH of enzyme solution was 5.8 and temperature was(26±1) ℃.Enzymolysis time was 8 h in dark.Further more,centrifuging the yield at speed of 1 000 rpm for 6 min.
