丹参抗LDL的脂质过氧化和脑缺血损伤的研究 被引量:6

摘要 给新西兰大白兔分别喂1% 胆固醇食(n= 19)和标准食(n= 15)共5 周,测定了血脂和低密度脂蛋白(LDL)中的脂质过氧化物(LPO)含量,并制成氧化型LDL(oxLDL),提取oxLDL中的脂质进行薄层层析,并制成脑缺血模型,观察丹参抗脑缺血损伤的作用。结果:胆固醇食组兔血清胆固醇(CH)、磷脂(PL)、游离脂肪酸(FFA)以及血浆和LDL中LPO 含量均显著高于标准食组(P< 0.01);胆固醇食兔LDL和Cu2+ 透析后的oxLDL中的脂质成分发生变化,X 带增加,FFA 带增高,而胆固醇酯(CE)带及TC带显著减少,而且丹参透析组脂质成分变化不显著,丹参组缺血后海马超微结构损伤较盐水组轻。表明,长期高胆固醇饮食可引起较多的LPO和oxLDL生成,从而加速动脉粥样硬化的形成;X 带可作为脂质过氧化的一个指标,丹参具有拮抗LDL 氧化的作用,这是其对抗脑缺血损伤的机制之一。 The blood lipids and lipid peroxide (LPO) were analysed in rabbits fed with 1% cholesterol in feeding ( n =19) and with standard feeding ( n =15) after 5 weeks of feeding.The plasma LDL was isolated and ox LDL was obtained by thin layer chromatography (TLC).Cerebral ischemia was induced by ligation of left common carotid artery.The antioxidation and antiischemia injury property of RSM was also observed with TLC and electron microscopy.The results showed that the plasma LPO,FFA,CH and PL levels in 1% cholesterol fed rabbits were much higher than those of rabbits with standard feeding( P <0 01).The lipid constituent changes in LDL in 1% cholesterolfed rabbits showed that the spot X was higher than that of standard feeding group.After dialysis with Cu 2+ ,the spot X and FFA were increased,but CE and TG bands were decreased,and the percentage of spot X in RSM dialysed was significantly lower than that of Cu 2+ dialysed LDL.The ultrastructural changes in hippocampus were more severe in saline treated group than those of RSM treated group.The results indicated that high cholesterol intake may induce disturbances of lipid metabolism and increase the formation of LPO,the spot X may be an important indicator for lipoperoxidation. RSM plays a potential role in antioxidartion of LDL and in the prevention of brain injury brain during cerebral ischemia.
