摘要
的 :阐明垂体腺苷酸环化酶激活肽 (PACAP)是否对血管平滑肌细胞 (SMC)具有抗损伤保护作用。方法 :以培养的猪肺动脉 SMC为实验对象 ,提取正常人血浆低密度脂蛋白 (L DL )造成细胞的高脂培养环境 ,光镜及电镜下观察 SMC在正常及高脂环境中受 PACAP作用所产生的形态改变。生化方法测定条件培养基中丙二醛 (MDA)的含量。结果 :1L DL 使 SMC的贴壁能力降低 ,细胞外形亦发生极大改变 ;电镜下可见 ,SMC由正常的收缩型变为合成型 ,且细胞膜破坏 ;2 L DL 使 SMC的 MDA产生显著增高 ;3 PACAP可明显减轻 L DL的上述损伤作用。结论 :PACAP可对抗 L DL对 SMC形态和功能的损伤 ,具有
Objective:Studying the cytoprotective effects of APCAP on vasular smooth muscle cell(SMC).Studying the cytoprotective effects of APCAP on vasular smooth muscle cell(SMC).Methods:The culture model of pulmonary artery's SMC of porcine were set up.Low density lipoprotein(LDL) from normal human fresh plasma were isolated and collected. The effects of APCAP on the SMC's morphology were observed by light and electronic microscope in normal and hyperlipid envronment. The concentration of MDA in the conditioned medium were measured by biochemical method. Results:①The SMCs were damaged by LDL:cell membrane were broken;cell junctions detached;contractile phenotype SMC transferred into synthetic phenotype.②LDL increased the content of SMC's MDA significantly. ③PACAP may partly counteract the morphologic injury of cells result from LDL.Conclusion:Conclusion:PACAP can prevent the morphologic damage of SMC from LDL.It is certainly that PACAP has vascular cytoprotective effects.
