摘要
[目的]探讨肝细胞性肝癌(hepatocellular carcinoma,HCC)中与硫酸乙酰肝素糖蛋白3(glypican3,GPC3)表达相关的Wnt信号通路及其他信号转导分子表达。[方法]通过实时荧光定量PCR方法检测GPC3过表达的肝癌细胞株以及不同肝病变组织样本中GPC3、Wnt、基质金属蛋白酶(MMPs)、内皮硫酸脂酶及其他信号分子表达。[结果]肝癌组织GPC3mRNA水平显着高于良性肝病变组织(P<0.05)。肝癌GPC3的表达与MMP2、14,内皮硫酸脂酶1、2,成纤维细胞生长因子及其受体FGF2、FGFR1,以及胰岛素样生长因子受体IGFR1R的表达具相关性。[结论]GPC3与MMPs、FGF和IGF结合,可能在肝癌的发展中起重要作用。
[Purpose] To investigate the association of glypican-3(GPC3) expression with Wnt and other signaling transduction molecules in hepatocellular carcinoma(HCC).[Methods] Expression of GPC3,Wnt,matrix metallo proteinases (MMPs),sulfatase1 (SULF1),SULF2 and other signaling molecules in GPC3-transfected HCC cells and in various liver tissue samples was analyzed by real-time reverse transcription-polymerase chain reaction.[Results] GPC3 mRNA was significantly higher in patients with HCC that in non-HCC subjects(P0.05).GPC3 expression was correlated with expressions of MMP2,MMP14,SULF1,SULF2,fibroblast growth factor receptor 1(FGFR1),insulin-like growth factor1 receptor(IGF1R).[Conclusion] GPC3,in conjunction with MMPs,FGF and IGF,might play an important role in the carcinogenesis of HCC.
