摘要
目的:探讨姬松茸多糖对糖尿病大鼠视网膜病变的保护作用及其作用机制。方法:将Wistar大鼠随机分为5组,A:正常对照组;B:模型组;C:8%姬松茸多糖组;D:4%姬松茸多糖组;E:2%姬松茸多糖组。8周后,琼脂糖凝胶电泳检测细胞凋亡,RT-PCR观察Bax基因、Survivin基因的表达。结果:与模型组比较,姬松茸多糖组视网膜细胞凋亡明显减少,Survivin表达上调,Bax表达降低。结论:姬松茸多糖对糖尿病大鼠视网膜组织保护作用的机制可能与抑制视网膜组织细胞凋亡及上调Survivin基因,降低Bax基因表达有关。
【Objective】 To investigate the effect and mechanism of Agaricus blazei Murill(ABM) polysaccharide on retina tissue of diabetic rats and its mechanism.【Method】 Wistar rats were randomly divided into 5 groups including diabetic model group,diabetic model treated with 8%ABM,4%ABM,2%ABM and normal control group.After 8 weeks,the number of apoptotic cells was detected by electrophoresis and the expression of Bax and Survivin gene was confirmed by RT-PCR.【Result】 Compared with diabetic model group,the number of apoptotic cells in retina tissue decreased and ABM could effectively decrease the level of gene Bax expression and improve the level of gene Survivin expression.【Conclusion】 ABM polysaccharide had protective effect on retina tissue of diabetic rats.The mechanism of protective effect of ABM polysaccharide on retina tissue was probably related to decreasing apoptosis and regulating Bax,Survivin gene expression.
