大豆异黄酮对运动去卵巢大鼠骨质疏松的影响 被引量:1

The influence of soybean isoflavone combined with exercise on ovariectomised rats osteoporosis

摘要 为了研究大豆异黄酮对运动去卵巢大鼠骨质疏松的预防作用,将60只3月龄SD大鼠按体重随机平均分成5组,即假手术组(SHAM,n=12),骨质疏松模型组(OVX,n=12),异黄酮组(ISO,n=12),运动+异黄酮组(ISO+E,n=12),雌激素组(E2,n=12)。所有动物分笼饲养,各组均接受腹腔手术。除SHAM组外,其他各组均切除卵巢。ISO组每天以30 mg/kg体重的剂量灌胃异黄酮,ISO+E组除了每天以30 mg/kg体重的剂量灌异黄酮外,每周跑台运动5次,E2组每天以0.1 mg/kg体重的剂量灌胃己稀雌酚。各组均自由饮水,喂食标准鼠类饲料,试验时间为3个月。3个月后,各组大鼠股动脉放血处死。留取子宫称重,剥离股骨,对股骨上端进行扫描电镜观察。主要结果如下。子宫指数:OVX组与SHAM组、E2组相比显著减少(P<0.01),ISO+E组、ISO组和OVX组比较子宫指数上升,ISO+E组与OVX组比较有显著差异(P<0.05);扫描电镜观察:OVX组的大鼠股骨头松质骨骨小梁与SHAM组、E2组相比有明显差异,OVX组的大鼠股骨头骨小梁结构不规则,骨小梁大多被吸收、破坏,骨小梁之间的连接丧失,骨小梁间的空隙有明显增大,小梁变细、变薄、穿孔、断裂,结构的完整性遭到破坏,骨小梁数目减少。ISO+E组与SHAM组、E2组结构相似,骨小梁数量多,且比较粗厚,间隙较小,结构完整。可见运动与大豆异黄酮联合作用可以抑制骨质疏松大鼠骨的微观结构退化,改善骨的微观结构,二者的联合作用具有抗骨质疏松的作用。 To study the joint effects of exercise and soybean isoflavone on ovariectomised rats osteoporo-sis,60 three-month-old female Sprangue-Dawley(SD) rats were randomly divided into 5 groups averagely accord-ing to weight,including sham operation group(SHAM,n=12),ovariectomised group(OVX,n=12),isoflavone treated group(ISO,n=12),combined group of exercise plus isoflavone(ISO+E,n=12) and estrogen treated group(E2,n=12).Rats in each group were kept in a separate cage,Except for the SHAM group,all groups of rats received celiac operation to have their ovaries removed.ISO group fed on isoflavone 30 mg/kg body weight eve-ryday;ISO+E group fed on isoflavone 30 mg /kg body weight everyday and ran on treadmill 5 times every week;E2 group fed on estrogen 0.1mg/kg body weight everyday.All rats drank water freely,and were fed on standard food.The experiment lasted for three months.After that,all rats were killed by cut femoral artery,stripping uterus and weighing,stripping femur,sem observation of top of the femur.The results are as follows.The uterus index: Compared with SHAM and E2 groups,OVX group uterus index significantly decreased(P 0.01),OVX group,ISO +E group and ISO group uterus index have significantly raised,there were significant differences in ISO + E group and OVX group(P 0.05).Scanning electron microscopy: there is obvious difference between OVX group femoral cancellous trabeculae and that of SHAM groups and E2 group,OVX groups femoral head bone trabeculae structure is irregular,trabeculae mostly were absorption,destruction,the connection between the bone trabeculae was disappeared,the gap between trabeculae increased obviously,trabecular became thin,perforation,fracture,the structural integrity was destruction,the numbers of bone trabeculae reduced.ISO +E,SHAM and E2 group structure is similar,have more trabeculae in number,trabeculae is thicker,clearance is smaller and complete structure.Therefore,exercise combined with isoflavone can inhibit bone microscopic structure degradation,im-prove bone microstructure.The joint effects can resist osteoporosis.
