电项针疗法对脑缺血再灌注大鼠ICAM-1及ICAM-1 mRNA表达的影响 被引量:7

The Effect of Electro-nape-acupuncture on the Expression of ICAM-1 and ICAM-1m RNA of Rats with Cerebral Ischemia-reperfusion

摘要 目的:观察电项针疗法对脑缺血再灌注后大鼠脑缺血半暗区细胞间粘附分子-1表达的影响。方法:运用栓线法制作局灶性脑缺血再灌注大鼠模型,随机将大鼠分成4组:假手术组、模型组、电体针组和电项针组,随机又将各组大鼠分为12 h、1 d、3 d和5 d共4个时点。通过HE染色的方法观察病理形态学的变化;应用免疫组织化学方法和原位杂交技术检测脑缺血半暗区ICAM-1和ICAM-1 mRNA的表达。结果:HE染色显示:与模型组相比,治疗组缺血半暗区的神经细胞肿胀程度减轻,胶质细胞增生,新生毛细血管和神经元的数量增多;电项针组明显于电体针组。治疗组可在早期抑制ICAM-1蛋白及其mRNA的表达(P<0.05,P<0.01)。同时间点电项针组与电体针组比较有显著性差异(P<0.05)。结论:电项针疗法可有效减轻神经元的损伤程度;电项针疗法可以下调再灌注后缺血区细胞间粘附分子-1蛋白及其mRNA的表达,从而达到防治脑缺血再灌注炎性损伤的目的。 Objective:To observe the effect of ICAM-1 by using mechanism of electro-nape-acupuncture on rats brain after acute cerebral infarction. Methods:Making rats model of brain ischemia by bolting the middle cerebral artery with monofilament.The rats were divided into four groups:fake group,pattern group,electro-acupuncture group and electro-nape-acupuncture group.The rats were randomly separated into twelve hours,one day,three days and five days.HE staining was used to observe the varies of histomorphology.To detect the expression of ICAM-1(mRNA) by using method of immunochemistryc and hybridization in situ. Results:Compared with electro-acupuncture group,electro-nape-acupuncture group grew more new neuron cells and capillary vessel,and the hurt in the cerebral ischemia lightened.But at the same time the electro-nape-acupuncture group was better than electro-acupuncture group in dominant. Conclusion:Electro-nape-acupuncture and electro-acupuncture group can help the numbers of renascence nerve cell reduce hurt.At the same time it can promote new horn cells and capillary vessel palingenesis in peri-infarct region and strengthen the blood supply in order to repair neural function.Electro-nape-acupuncture can lessen neurologic impairment,decrease cell apoptosis,restrain ICAM-1(mRNA)expressions,which indicates that the action on repressing cell apoptosis is related to the regulation on ICAM-1(mRNA)expression.
